After defeating Ruin Guard, you need to wait for the server reset the next day for it to respawn.

Ruin Guards, just like other Ruin enemies, are resistant to physical attacks. Use Elemental Attacks or Elemental Reactions to defeat them.

Ruin Guards have two weak points: The eye on their head and the light on their back. Shoot any of the two twice and it will stun it!
