There is a limited number of NPC that you can challenge per level. You can find and challenge available NPCs with the help of Casket of Tomes.

However, you can't challenge all NPCs immediately. Some NPCs are level-locked, meaning you have to be at a certain level first before they can be challenged.

Tavern Challenge Friendly Fracas & Serious Showdown
Similar to inviting characters mentioned above, you cannot gain EXP from Tavern Challenges so it is ok to skip it if gaining EXP is your priority.
