If you use Exp. Candy to level up a Pokemon to Level 100 all at once, a Pokemon with two evolutions will evolve only once. You may use Rare Candy after evolving to Level 100 as well BUT it is recommended to separate Exp. Candy into 2 uses, so you don't have to waste Rare Candy.
