Huge Rupee Location
House of Skulltula

Collect all 100 Gold Skulltulas, then speak to the Cursed Rich Man in the House of Skulltula to receive a Huge Rupee as a reward. You can claim the Huge Rupee repeatedly by exiting and re-entering the House of Skulltula, then speaking to the Cursed Rich Man again.

Defeat Skull Kid
Complete the Trading Quest, then enter the Lost Woods and turn left to find Skull Kid. Approach Skull Kid and attack until they are defeated to obtain an orange Huge Rupee. To collect the Huge Rupee repeatedly, exit and re-enter the Lost Woods to respawn Skull Kid.
Without Completing Trading Quest

You can obtain a Huge Rupee without completing the Trading Quest by defeating the two Skull Kids instead. However, using projectile attacks or taking too long to defeat them will cause the Skull Kids to despawn. To collect the Huge Rupee repeatedly, exit and re-enter the Lost Woods to respawn the Skull Kids.
