Points of No Return & Missable Content
Village: Can't Enter Village After Entering Stronghold
While you will move to and from the Village quite often between main missions, you can never return to the Village once you've entered the Stronghold. All uncollected items are thus lost.

Castle Dimitrescu is another point of no return. Once you get all 4 masks & get the dagger, you can no longer go back inside the castle after your boss fight with Lady Dimitrescu.

Exiting a boss' room/level locks it permanently, blocking access to the items stashed within. If you leave the room/level, all uncollected attachments, ingredients, items inside will be lost, so make sure to explore before you leave.
Locations & Setting GuideWeapon Upgrades Are Permanent
The choice to upgrade your weapon is permanent. You cannot redo the choice and get your materials back even if you feel the upgrade wasn't worth it.

In RE8, Your bullets and healing items will be limited throughout the game. The resources you expend might not be replenished even if you move on to the next challenge, so if you can get past a section without fighting, by all means!

Missable Fish: Reservoir

 | Can be accessed after you get out of the boat. After disembarking, do not go to the hut immediately and turn right |
 | Can be accessed after defeating Moreau, after the water has been drained |
Be careful with these locations as the fishes in the Reservoir are missable. You will not have access to them again once you take the elevator to leave the Reservoir.
