Once you finish a mission or quest, it will not be replayable. It will remain that unless you redo the whole story with a new save or in True Vault Hunter Mode.

Mission Reward Choices Are Permanent
Sometimes, completing a mission will reward you with a choice between useful items. Make sure to choose wisely as your pick will be irreversible. This also include optional bonuses.
Character Choices & Points of No Return
Character Choice Is Permanent
Choosing one of the four Vault Hunters during the first part of the game is permanent. Make sure that you are happy with the abilities and skills of the Vault Hunter before making your final choice!

Cannot Lower Levels
Once you've leveled up, there's no going back from there. You cannot lower your level to do lower the difficulty or get on the same level of a low level mission.
You Can Respec Skill Points
If you are unhappy with your Vault Hunter's current build, you will be able to respec your skill points at any Quick Change station for a price. Keep in mind that respeccing skill points requires you to pay money each time.

Investing in Guardian Rank is Permanent
Once you've used up your Guardian Rank to buy a buff, it cannot be reversed. There is no respeccing when it comes to Guardian Ranks!

Purchasing backpack upgrades and other things from Crazy Earl is a done deal. You won't even get an option to return your purchased item.
Spent SHIFT Keys are Gone
SHIFT Keys will be taken immediately from your inventory once you've used them to open the Treasure Chest in Sanctuary III. Didn't like what you got? Too bad because you won't get your SHIFT Key back.

Sold Items from your inventory can be repurchased for the same price in the Vending Machine. However, once you leave the vending machine menu, the item will be gone forever!
