Defeat Lady Butterfly At The Hirata Estate
When you defeat Lady Butterfly at the Hirata Estate, you'll receive this item as one of the rewards. One of the most difficult bosses in the game, you need to combine bringing down her HP & Posture to defeat her.

There are 2 ways for you to use the Sakura Droplet. You can show it to Kuro at the Ashina Castle or the Divine Child of Rejuvenation.
Talk To Isshin Ashina To Activate Kuro
In order to be able to show Kuro the Sakura Droplet, you need to talk to Isshin Ashina in Ashina Castle first. He'll talk to you about the Sakura Droplet which will allow you to show it to Kuro.
Get Mortal Blade To Activate Divine Child Of Rejuvenation
You must first retrieve the Mortal Blade from the Divine Child of the Rejuvenation before you can give her the Sakura Droplet. You just need to talk to her to open the option of giving her the item.
What Is The Sakura Droplet?
Item That Activates Additional Resurrection Node
Alone the Sakura Droplet is just another item. However, if you give it to either Kuro or the Divine Child, you will activate an additional resurrection node, from 2 to 3.
