Differences Between Each Class
Tank, Healer, and DPS
Each character role or class caters to one specific archetype. Essentially, each class represents a Tank, a DPS, and a Support. Even though each character has access to unique abilities, their kit will typically be themed around doing damage, supporting allies, or increasing survivability.
Vanguard
Vanguards are the equivalents of tanks in most other games. These characters have a high HP pool and have defensive abilities that make themselves or their allies more durable.
How to Play Vanguards
As the tank, your main task will be to frontline for your team. Initiating the fight, absorbing damage, and shielding allies from attacks should be your priority whenever you're about to skirmish.
When in doubt, just make sure to stand in front of everyone else to pull attention towards your character. This will typically provide your allies with openings to punish the opposing team.

Duelist
Duelists take up the the job of DPS specialists. Characters that belong to this category are oriented towards offense, allowing them to deal significant amounts of damage in short periods of time.
How to Play Duelists
Duelists are opportunists in essence. These characters excel at dealing large amounts of damage, but only have small HP pools as a trade-off.
Leverage your high damage output to pick-off enemy Strategists, or opposing Duelists to gain an advantage towards your objectives.

Strategists
Strategists are the supports of Marvel Rivals. Characters that belong to this category often have abilities that focus on utility. Things like heals, buffs, and debuffs are common themes for these characters.
How to Play Strategists
Strategists vary quite substantially from one another in Marvel Rivals, however, the key aspect in playing as a Strategist effectively is timing.
Selecting the perfect moment to heal, shield, resurrect, or buff your allies is of utmost importance, and can turn the tide of the match when timed precisely.
